Pharmacokinetics Basics: Half-Life, Clearance & Steady State

First-order elimination, half-life and k from two levels, volume of distribution, clearance and AUC with the trapezoid rule, accumulation to steady state, time to plateau, infusions and the effect of kidney function, all in a live spreadsheet that checks every cell. Placeholder drugs and illustrative parameters only, not clinical guidance.

What you'll build

What you will be able to do

Each lab ends with a concrete result that you build yourself and the checker confirms, cell by cell.

  • Model the plasma concentration of placeholder Drug K after an IV bolus over 24 hours, the share that remains, the time to fall to a reference level, and chart the decay curve.
  • Estimate the elimination rate constant, the half-life and the back-extrapolated starting concentration of Drug K for six subjects from two blood samples each.
  • Calculate the apparent volume of distribution of Drug K for five subjects, scale it per kilogram and apply the loading-dose formula with bioavailability as a calculation exercise.
  • Calculate the area under the concentration-time curve of Drug K with the trapezoid rule, add the extrapolated tail and derive the clearance, then compare it with k × Vd.
  • Follow peaks and troughs of Drug K over ten repeated IV doses, compare them with the steady-state formula and chart the build-up to the plateau.
  • Compare five placeholder drugs: elimination rate constant, accumulation ratio for their dosing interval and the time to reach 95% of steady state.
  • Calculate the steady-state concentration of a constant-rate Drug K infusion, its elimination rate constant, the concentration over 48 hours and the time to reach 90% of the plateau.
  • Estimate how reduced kidney function lowers the total clearance and lengthens the half-life of Drug K for six practice patients, and flag the cases that need review.

About this course

Pharmacokinetics is built on a handful of exponential equations, and they make far more sense once you calculate them. This course is a study aid for pharmacy, medical and life-science students. You model first-order elimination in a time table and chart, derive half-life and the rate constant from two concentrations and relate volume of distribution to the loading-dose formula. Clearance and AUC are calculated with the trapezoid rule. Repeated dosing shows accumulation towards steady state, with the accumulation ratio and the four-to-five half-life rule, and a continuous infusion approaches its plateau. A final lab demonstrates an illustrative proportional adjustment for renal function with safety flags. For learning only; never use it for real dosing decisions.

Questions about this course

Is this a subscription?

No. You pay $9.99 once for this course. There is no subscription and nothing renews automatically. Prices are in US dollars; taxes may be added where required.

What exactly do I get?

All lessons and labs of this course, the final exam and the certificate, including later updates to the course, for as long as Tutorvo offers it. Access is personal and tied to your account.

Can I get a refund?

Yes. If you are not satisfied, email hello@tutorvo.com within 14 days of buying and we refund the course in full via Stripe. The guarantee does not apply once a certificate for the course has been issued to you, or where there are signs of abuse.

When can I start?

Right after payment. Access starts immediately at your request; for consumers in the EU/EEA the statutory right of withdrawal therefore ends once access has begun, while our 14-day money-back guarantee applies regardless.

Do I need Excel or other software?

No. The labs run in a spreadsheet inside your browser on a laptop, tablet or phone. Function names and syntax follow Excel.

Is the certificate an official qualification?

No. It confirms that you completed all lessons of the course and passed its final exam. It is not an official, state-recognised or professional qualification. The name is taken from your profile when the certificate is issued.
